Major service or electrical tune?
Tune up is such a non-specific term.

Valve job, install clutch, change bushings, change all fluids. At least that's what's waiting for me. Told the wife it just needs a little work.
Anyone can drive a new car.

Just remember that when she tells you she is going to town to do a "little" shopping..........hehehehehehe

When I first got my Z about year and a half ago, the local parts store always had to order me one in. Now that I have two Z's, they have started stocking L28 fuel filters again. lol
Not as good as the Courtesy ones though, I try to order a few from them along with other tune up components as necessary. My local Nissan dealerships are useless for original Z parts.
Have been taking everyone's advise and replacing the G3's on a regular basis, sure is cheaper that way and you find out real quick what is floating around in your tank.

1987 Toyota Supra Turbo 5-speed. Belongs to me until my friend get's back from Iraq.

Not one person has mentioned a brake inspection... They at least should be checked.

What kind of vehicle? How old? What kind of use?
There are many ideas of major or minor tune-up - all jokes aside.
If it's Zcars do Search & Articles - people like Blue, Tony D. and many other wonderful people have specific advice and articles on many aspects of tune-up.
Much of this applies to other cars.
Be much more specific in your questions.
